There's one I've been thinking of. One with Trolls which are immune to fire and acid and require necromancy to kill, (so there are also "Ju-Ju Trolls") and the Darklord is a human-on-one half, troll on the other Necromancer/Psion.

Sounds good. Now, does this new subspecies have a unique culture or are they just a new variety that acts like the common one?

I was thinking I might have the darklord hire/employ the PC's to rid a place of these trolls, only to discover it was him who designed them in the first place. I figured he might give them some item which could help kill them, maybe a severed finger which acts a rod of finger of death or something. Or a sword that drains life from them. Maybe a sphere of annihilation.

Also, there was a spell in the 2E wizards handbook called black mantle, which prevented a troll from regenerating while it was in effect, but I forget what happens once that spell wears off, if the troll reaches 0 while the spell is in effect, drops to the ground, and the spell ends, does it regenerate from there, if which case it wouldn't be that useful, just delaying his regen for a few rounds.

Would disintegrate kill a troll, if you got the whole troll? I'd say yes if it doesn't leave any ash/dust, which could regenerate.

Is there a particular reason this part-troll has to be a darklord? This all sounds to me like something a plain ol' garden variety bad-guy could do.

I could say it was some kind of regen experiment gone awry, he wanted trolls regen but instead accidently made himself a troll hybrid.

Well I do hope you plan to elaborate on this Darklord a bit. Remember that Dark Lords aren't just high-level villains to save the princess from... they are deep, tragic figures that are trapped by their own desires. In fact, a domain is more based around its darklord than vice versa, as the domain is mostly a reflection of the dark lords personality.

The meat and potatoes of the story behind why he is a darklord should revolve around the experiment then. It would be more in line with being cursed or punished if he was using trolls or troll parts in his experiment. Of course, he could have a pocket domain instead of a full sized one.
